In this new paper (https://doi.org/10.3390/molecules24142557), we established how to choose the analysis criteria to retrieve accurate FRET efficiencies from single-molecule fluorescence experiments and to prevent collecting data that does not purely originate from single molecules.
Interestingly, until today such a systematic assessment of the underlying meaning of single molecule burst analysis has not been taken, and the choice of burst analysis parameter values was somewhat arbitrary. This new paper makes that choice physically meaningful.
